Disclosed is a method for detecting 5-methylcytosine in genomic DNA samples. First, genomic DNA from a DNA sample is chemically reacted with a reagent, 5- methylcytosine and cytosine reacting differently. The pre-treated DNA is then ampified with primers from a different sequence using a polymerase. In the following step, the amplified genomic DNA is hybridised to an oligonucleotide array and PCR products are obtained which must be provided with an identifying mark. Alternatively, the PCR products can be extended in a Primer Extension Reaction, the extension products also being provided with an identifying mark. The last step involves examining the extended oligonucleotides for the presence of the identifying mark.
L'invention concerne un procédé servant à mettre en évidence la présence de 5-méthylcytosine dans des échantillons d'ADN génomiques. On fait d'abord réagir chimiquement un ADN génomique provenant d'un échantillon d'ADN avec un réactif, la 5-méthylcytosine et la cytosine réagissant différemment. On amplifie ensuite l'ADN préalablement traité en utilisant une polymérase à l'aide d'amorces de séquence différente. A l'étape suivante, on hybride l'ADN génomique amplifié sur un jeu ordonné d'oligonucléotides et on obtient des produits PCR qui doivent être pourvus d'un repère. On peut également allonger les produits PCR dans une réaction d'extension d'amorce, les produits d'allongement étant également munis d'un repère. Lors de la dernière étape, on examine les nucléotides allongés afin de déterminer si le repère est présent.
