A method for determining the absolute cell count of an identifiable cell sub-population contained in a sample, the method which comprises the steps of introducing into an aliquot of the biological sample a pre-determined amount of a fluid reagent which comprises a known concentration of fixed and labeled cells and using flow cytometry to determine a count of the cells of the cell sub-population, and to determine a count of the labeled cells. The cell sub-population count and the labeled cell count is used to determine an absolute cell count of cell sub-population.
