The present invention relates to tricyclic derivatives or pharmaceutically acceptable salts thereof, their preparations and pharmaceutical compositions containing them. More precisely, the present invention relates to tricyclic derivatives as colchicine derivatives, pharmaceutically acceptable salts thereof, their preparations and pharmaceutical compositions containing them. Tricyclic derivatives of the present invention show very powerful cytotoxicity to cancer cell lines but were much less toxic than colchicine or taxol, confirmed through animal toxicity test. Tricyclic derivatives of the invention also decrease the volume and weight of a tumor and have a strong angiogenesis inhibiting activity in HUVEC cells. Thus, tricyclic derivatives of the present invention can effectively be used as an anticancer agent, anti-proliferation agent and an angiogenesis inhibitor.
La présente invention se rapporte à des dérivés tricycliques ou à des sels pharmaceutiquement acceptables de ces dérivés tricycliques, à leurs préparations et à des compositions pharmaceutiques les contenant. Plus précisément, la présente invention se rapporte à des dérivés tricycliques tels que des dérivés de colchicine, à des sels pharmaceutiquement acceptables de ces dérivés, à leurs préparations et à des compositions pharmaceutiques les contenant. Les dérivés tricycliques de la présente invention présentent une cytotoxicité très puissante vis-à-vis des lignées cellulaires cancéreuses mais il s'avère moins toxique que la colchicine ou le taxol, ce qui est confirmé par un essai de toxicité sur des animaux. Les dérivés tricycliques de cette invention réduisent également le volume et le poids d'une tumeur et ils possèdent une forte activité d'inhibition de l'angiogenèse dans des cellules HUVEC. Ainsi, les dérivés tricycliques de la présente invention peuvent être utilisés efficacement en temps qu'agents anticancéreux, agents antiprolifération et inhibiteurs de l'angiogenèse.
