A process for providing regiospecific and highly stereoselective synthesis of 9-.beta. anomeric purine nucleoside analogs is described. The introduction of the sugar moiety on to 6-(azolyl)-substituted purine bases is performed so that highly stereoselective formation of the .beta. anomers of only the 9 position regioisomers of the purine nucleoside analogs (either D or L enantiomers) is obtained. This regiospecific and stereoselective introduction of the sugar moiety allows the synthesis of nucleoside analogs, and in particular 2'-deoxy, 3'-deoxy, 2'-deoxy-2'-halo-arabino and 2',3'-dideoxy-2'-halo-threo purine nucleoside analogs, in high yields without formation of the 7-positional regioisomers. Processes for providing novel 6-(azolyl)purines for the regiospecific and highly stereoselective synthesis of 9-.beta. anomeric purine nucleoside analogs are described. The compounds are drugs or intermediates to drugs.

L'invention concerne un procédé permettant d'obtenir une synthèse régiospécifique et hautement stéréosélective d'analogues de nucléosides de purines anomériques 9-ß. L'introduction de la fraction de sucre sur des bases de purines 6-(azolyl)-substituées est effectuée de manière qu'une formation hautement stéréosélective des anomères ß d'uniquement les régiosomères en position 9 des analogues de nucléosides de purines (soit des enantiomères D ou L) soit obtenue. Cette introduction régiospécifique et stéréosélective de la fraction de sucre permet d'obtenir la synthèse des analogues de nucléosides, notamment des analogues de nucléosides de purines 2'-désoxy, 3'-désoxy, 2'-désoxy-2'-halo-arabino et 2',3'-didésoxy-2'-halo-thréo, à des rendements élevés sans formation des régiosiomères en position 7. L'invention concerne également des procédés permettant de préparer des purines 6-(azolyl) destinées à la synthèse régiospécifique et hautement stéréosélective d'analogues de nucléosides de purines anomériques 9-ß. Les composés sont des médicaments ou des intermédiaires aux médicaments.
