The present invention relates to a novel method for synthesizing peptides in solution, the method comprising: a) treating a peptide or amino acid derivative, which has an .alpha.-amine function blocked with a urethane group of the formula R1-SO2-CH2CH2-O-CO-, wherein R1 is aryl, with an excess of an aliphatic secondary amine in an organic solvent to provide the release of the free .alpha.-amine function of said amino acid or peptide derivative and the formation of a tertiary amine adduct between said secondary amine and the liberated vinyl compound of the formula R1-SO2-CH2=CH2; b) removing the solvent and the excess of said secondary amine; c) contacting thus formed mixture of the N.alpha.-deprotected amino acid or peptide derivative and said tertiary amine adduct with subsequent peptide or amino acid derivative N.alpha.-protected with the above indicated or another urethane group under conditions providing the formation of a peptide bond between the free .alpha.- amine function of the N.alpha.-deprotected amino acid or peptide derivative and an .alpha.-carboxylic function of the subsequent N.alpha.-protected peptide or amino acid derivative; d) separating the newly formed peptide from the reaction mixture; e) repeating procedures as set forth above until the desired polypeptide is obtained.

Cette invention se rapporte à un nouveau procédé pour la synthèse de peptides en solution, ce procédé consistant : (a) à traiter un dérivé de peptide ou d'acide aminé, qui possède une fonction .alpha.-amine bloquée par un groupe uréthane de la formule R?1¿-SO¿2?-CH¿2?CH¿2?-O-CO-, où R?1¿ représente aryle, avec une quantité excédentaire d'une amine secondaire aliphatique dans un solvant organique, pour produire la libération de la fonction .alpha.-amine libre de ce dérivé d'acide aminé ou de peptide et la formation d'un produit d'addition à base d'amine tertiaire entre la seconde amine secondaire et le composés vinyle libéré de la formule R?1¿-SO¿2?-CH¿2?=CH¿2 ?; (b) à retirer le solvant et la quantité excédentaire de l'amine secondaire ; (c) à mettre en contact le mélange ainsi formé du dérivé d'acide aminé ou de peptide déprotégé en position N¿.alpha.? et du produit d'addition à base d'amine tertiaire avec un dérivé de peptide ou d'acide aminé ultérieur protégé en position N¿.alpha.? par le groupe uréthane indiqué ci-dessus ou par un autre groupe uréthane, dans des conditions entraînant la formation d'une liaison peptide entre la fonction .alpha.-amine libre du dérivé d'acide aminé ou de peptide déprotégé en position N¿.alpha.? et une fonction .alpha.-carboxylique du dérivé de peptide ou d'acide aminé ultérieur protégé en position N¿.alpha. ?; (d) à séparer le peptide nouvellement formé du mélange de réaction ; (e) et à répéter les procédures décrites ci-dessus, jusqu'à obtenir le polypeptide souhaité.
