A procedure for the preparation of the radioactive tracer 3'-deoxy-3'-[18F]fluorothymidine ([18F]FLT) comprises the following steps: a) preparation of anhydrous [18F]F- fluoride; b) labelling of a precursor, for example 3-N-Boc-1-[5'- 0- ( 4, 4' -dimethoxytrityl ) -3' -O-nitrophenylsulf onyl-2' - deoxy-.beta.-D-lyxofuranosyl ] thymidine; c) hydrolysis of the mixture obtained by means of the previous steps; d) purification of the mixture to obtain the radioactive tracer [18F]FLT. In this procedure steps a), b) and c) are carried out by means of known methods. In step d) the mixture obtained after step c) is purified by means of the passage through a ventilated filter and one or more cartridges of the cationic exchange type (e.g. a Chromafix PS-H+), one or more cartridges of the reversed phase type, and finally a passage through cartridges either of the alumina N type, or diol cartridges, or anionic exchange cartridges or a combination thereof. Furthermore, the mixture which is present in the cartridges is eluted with H2O for injection, whereby the resulting product is forwarded to a container or ampoule for the collection of [18F]FLT.
La présente invention a pour objet une procédure de préparation du traceur radioactif 3'-désoxy-3'-[18F]fluorothymidine ([18F]FLT) qui comprend les étapes suivantes : a) préparation de fluorure anhydre [18F]F-; b) marquage d'un précurseur, par exemple, le 3-N-Boc-1-[5'-0- (4,4'-diméthoxytrityl)-3'-O-nitrophénylsulfonyl-2'-désoxy-.beta.-D-lyxofuranosyl]thymidine; c) hydrolyse du mélange obtenu au moyen des étapes précédentes; d) purification du mélange pour obtenir le traceur radioactif [18F]FLT. Dans cette procédure, les étapes a), b) et c) sont réalisées au moyen de procédés connus. Dans l'étape d), le mélange obtenu après l'étape c) est purifié au moyen du passage à travers un filtre ventilé et d'une ou plusieurs cartouches de type échangeur de cations (par exemple, une Chromafix PS-H+), d'une ou plusieurs cartouches du type phase inversée et finalement, un passage à travers des cartouches soit de type alumine N, soit des cartouches de diol soit des cartouches d'échange anionique soit d'une combinaison de celles-ci. En outre, le mélange qui est présent dans les cartouches est élué avec H2O pour l'injection et cette élution dirige le produit résultant vers un récipient ou une ampoule pour le recueil de la [18F]FLT.
