The present invention relates to the new 3-decladinosyl derivatives of 9a-N- carbamoyl~and 9a-N-thiocarbamoyl-9-deoxo-9-dihydro-9a-aza-9a-homoerythromycin A of the general formula (I), their pharmaceutically acceptable addition salts with inorganic or organic acids and their hydrates, Wherein R1 individually stands for hydrogen or together with R2 stands for double bond, R2 individually stands for hydrogen, hydroxyl or a group of the formula (II), wherein Y individually stands for monocyclic aromatic ring, unsubstituted or substituted with groups which are selected independently from halogen, OH, OMe, NO2, NH2 or R2 together with R3 stands for ketone or together with R1 stands for double bond, R3 individually stands for hydrogen or together with R2 stands for ketone or together with R4 stands for ether, R4 individually stands for hydroxyl, a OCH3 group or together with R3 stands for ether, R5 individually stands for C1-C4 alkyl group, C2-C4 alkenyl group, -(CH2)m Ar, wherein Ar individually stands for phenyl or phenyl substituted with one or two groups which are selected independently from halogen or halogen alkyl, and m is 0-3, R6 individually stands for hydrogen or hydroxyl protected group, X stands for oxygen or sulphur, to intermediates for synthesis of other macrolide compounds with antibacterial activity, to the process for their preparation, to their pharmaceutically acceptable addition salts with inorganic or organic acids and their hydrates, to the process for the preparation of pharmaceutical compositions, as well as the use of pharmaceutical compositions for treating bacterial infections.
L'invention concerne des dérivés de 3-décladinosyle de la 9a-N-carbamoyl- et de la 9a-N-thiocarbamoyl-9-désoxo-9-dihydro-9a-aza-9a-homoérythromycine A de formule générale (I), des sels d'addition acceptables sur le plan pharmaceutique avec des acides inorganiques ou organiques et les hydrates de ceux-ci, dans laquelle R¿1? représente de manière individuelle l'hydrogène ou conjointement avec R¿2? une liaison double, R¿2? désigne de manière individuelle l'hydrogène, l'hydroxyle ou un groupe de formule (II), dans laquelle Y représente individuellement un noyau aromatique monocyclique, non substitué ou substitué avec des groupes sélectionnés de manière indépendante parmi l'halogène, OH, OMe, NO¿2?, NH¿2? ou R¿2? conjointement avec R¿3? représente la cétone ou conjointement avec R¿1? désigne une liaison double, R¿3? représente individuellement l'hydrogène ou conjointement avec R¿2? représente la cétone ou conjointement avec R¿4? désigne l'éther, R¿4? représente de manière individuelle l'hydroxyle, un groupe OCH¿3? ou conjointement avec R¿3? désigne l'éther, R¿5? représente de manière individuelle le groupe alkyle en C¿1?-C¿4?, le groupe alcényle en C¿2?-C¿4?, -(CH¿2?)¿m? Ar, où Ar représente de manière individuelle le phényle ou un phényle substitué avec un ou deux groupes sélectionnés de manière indépendante parmi l'halogène ou l'halogène alkyle et m prend une valeur comprise entre 0-3, R¿6? représente de manière individuelle l'hydrogène ou un groupe protégé d'hydroxyle et X représente l'oxygène ou le soufre. L'invention concerne également des intermédiaires destinés à la synthèse d'autres composés de macrolide avec une activité antibactérienne, le procédé de préparation de ceux-ci, les sels d'addition acceptables sur le plan pharmaceutique de ceux-ci avec des acides inorganiques ou organiques et les hydrates de ceux-ci, le procédé de préparation des compositions pharmaceutiques, ainsi que l'utilisation de celles-ci dans le traitement d'infections bactériennes.
