The present invention relates to methods and compositions for increasing the production of high titer stocks or recombinant AAV (rAAV) through regulation of expression of the AAV REP and CAP proteins. The methods and compositions of the invention are based on the observation that the low level expression of the AAV REP 78/68 protein increases the production of AAV viral capsid protein and efficiency of packaging resulting in production of higher titer recombinant viral stocks. The invention encompasses recombinant AAV vectors that direct the expression of AAV REP and CAP proteins and the use of such vectors for the production of novel stable cell lines capable of generating high titer rAAV vectors. The invention provides methods for regulating the expression of the AAV REP 78/68, REP 40/52 and CAP gene at the transcriptional and post-transcriptional level.
La présente invention a trait à des procédés et compositions servant à augmenter la production de réserves à titre élevé de virus AAV recombinants (rAAV) en régulant l'expression des protéines rep et CAP du virus AAV. Les procédés et compositions de cette invention sont fondés sur l'observation que l'expression à bas niveau de la protéine rep 78/68 du virus AAV augmente la production de la protéine capsidiale du virus AAV et l'efficacité d'encapsidation, ce qui entraîne la production de réserves de virus à titre élevé. L'invention concerne aussi les vecteurs du virus AAV recombinant qui dirigent l'expression des protéines rep et CAP du virus AAV ainsi que l'utilisation de ces vecteurs dans la fabrication de nouvelles lignées cellulaires stables, capables de générer des vecteurs de rAAV à titre élevé. Enfin, l'invention concerne des procédés de régulation de l'expression du gêne rep 78/68/, rep 40/52 et CAP de l'AAV au niveau transcriptionnel et post-transriptionnel.
