The present invention provides a method for isolating cyclosporin by adsorption chromatrography. The method comprises the steps of: (a) loading a crude solution containig a cyclosporin onto adsorption chromatography column containing an adsorption resin, (b) eluting the cyclosporin from the adsorption chromatography column, and (c) collecting the eluted cyclosporin. The step of eluting the cyclosporin is carried out by passing an eluting solvent through the column, which comprises a hydrophilic component and a lipophilic component, wherein the cyclosporin is separately eluted from other compounds by providing a hydrophilic/lipophilic balance in the eluting solvent that selectively desorbs the cyclosporin from the column.
Cette invention se rapporte à un procédé pour isoler de la cyclosporine par chromatographie par adsorption. Ce procédé consiste: (a) à charger une solution brute contenant une cyclosporine dans une colonne de chromatographie par adsorption contenant une résine d'adsorption; (b) à séparer par élution la cyclosporine de la colonne de chromatographie par adsorption; et (c) à recueillir la cyclosporine éluée. On effectue l'étape d'élution de la cyclosporine en faisant passer à travers la colonne un solvant d'élution, lequel contient un constituant hydrophile et un constituant lipophile, la cyclosporine étant séparée par élution des autres composés grâce à un équilibre hydrophile/lipophile dans le solvant d'élution, entraînant une désorption sélective de la cyclosporine à partir de la colonne de chromatograhie.
