The invention relates to neuromodulatory oligomers of the amyloid-.beta.(1-42) protein, a particular production method, by means of which the oligomer can be obtained in a reproducible manner at high yield, the use of the oligomers as diagnostic and therapeutics agents, for the generation of oligomer-specific antibodies and for the discovery of substances which can interact with the oligomers and in the formation thereof. Corresponding methods for the production of the antibodies and for discovery of the substances are also disclosed as are the antibodies themselves and the use of the antibodies or substances as diagnostic and therapeutic agents. The invention further relates to derivatives of the oligomers and oligomers based on abbreviated forms of the amyloid-.beta.(1-42) proteins, the production and use thereof.
La présente invention concerne des oligomères neuromodulants de la protéine .beta.(1-42) amyloïde, un procédé de fabrication spécifique permettant de fabriquer les oligomères de façon reproductible, à un rendement élevé, ainsi que l'utilisation des oligomères en tant qu'agents diagnostiques et thérapeutiques, pour la production d'anticorps spécifiques aux oligomères, et la découverte de substances pouvant interagir avec les oligomères et leur formation. L'invention concerne également des procédés de fabrication des anticorps ou de découverte desdistes substances, ainsi que les anticorps et l'utilisation desdits anticorps ou substances en tant qu'agents diagnostiques et thérapeutiques. L'invention concerne par ailleurs des dérivés des oligomères selon l'invention, notamment des oligomères à réticulation transversale, et des oligomères à base de formes raccourcies de la protéine .beta.(1-42) amyloïde, et la fabrication et l'utilisation de ces oligomères.
