Layered pharmaceutical composition suitable for oral use in the treatment of diseases where absorption takes place over a large part of the gastrointestinal tract. The composition comprising A) a solid inner layer comprising i) an active substance, and ii) one or more disintegrants/exploding agents, one of more effervescent agents or a mixture thereof. the solid inner layer being sandwiched between two outer layers B1) and B2), each outer layer comprising iii) a substantially water soluble and/or crystalline polymer or a mixture of substantially water soluble and/or crystalline polymers, the polymer being a polyglycol in the form of one of a) a homopolymer having a MW of at least about 100,000 daltons, and b) a copolymer having a MW of at least about 2,000 daltons, or a mixture thereof, and iv) an active substance, which is the same as in said solid inner layer A), and layer A being different from layer B, the layered composition being coated with a coating C) that has at least one opening exposing at least one surface of said outer layer, the coating being substantially insoluble in and impermeable to fluids and comprising a polymer, and the composition having a cylindrical form optionally with one or more tapered ends, wherein the ratio between the surface area of one end surface of the cylinder and the length of the cylinder is in a range of from 0.02 to 45 mm.
L'invention concerne une composition pharmaceutique stratifiée appropriée pour une utilisation orale dans le traitement de maladies où l'absorption a lieu sur une grande partie du tractus gastro-intestinal. La composition comprend A) une couche interne solide comprenant i) une substance active, et ii) un ou plusieurs désintégrants/agents explosants, un ou plusieurs agents effervescents ou un mélange de ceux-ci, la couche interne solide étant prise en sandwich entre deux couches externes B1) et B2), chaque couche externe comprenant iii) un polymère sensiblement soluble dans l'eau et/ou cristallin ou un mélange de polymères sensiblement solubles dans l'eau et/ou cristallins, le polymère étant un polyglycol sous la forme de l'un parmi a) un homopolymère ayant une masse moléculaire d'au moins environ 100000 daltons, et b) un copolymère ayant une masse moléculaire d'au moins environ 2000 daltons, ou un mélange de ceux-ci, et iv) une substance active, qui est identique à celle dans ladite couche interne solide A), et la couche A étant différente de la couche B, la composition stratifiée étant revêtue d'un revêtement C) qui a au moins une ouverture exposant au moins une surface de ladite couche externe, le revêtement étant sensiblement insoluble dans et imperméable aux fluides et comprenant un polymère, et la composition ayant une forme cylindrique avec facultativement une ou plusieurs extrémités effilées, le rapport entre l'aire de surface d'une surface d'extrémité du cylindre et la longueur du cylindre se trouvant dans une plage de 0,02 à 45 mm.
