A novel strategy for monitoring both the expression and/or localization of a transgene in a mammal is disclosed. A marker peptide and/or sodium iodide symporter is genetically fused to either the N-terminus or C-terminus of the product of a transgene through a linker peptide which bears the recognition sequence of a host cell protease. Expression of the transgene results in release of the marker peptide into extracellular body fluid of the mammal in proportion to the amount of transgene product. The level of the released marker peptide serves as an indicator of the level of transgene expression. Expression of the transgene confers the activity of the sodium iodide symporter (NIS) to a host cell which expresses the transgene. Subsequent administration of labeled iodine results in transport of the labeled iodine into the cell bearing the NIS, which can then be localized and measured using standard imaging techniques. The system is particularly useful for monitoring the expression and localization of therapeutic transgenes and production of the therapeutic gene products.

L'invention concerne une nouvelle technique de surveillance de l'expression et/ou de la localisation d'un transgène chez un mammifère. Un peptide marqueur et/ou un symporteur iodure de sodium est génétiquement fusionné à l'extrémité N-terminale ou C-terminale du produit d'un transgène par un peptide lieur qui porte la séquence de reconnaissance d'une protéase de cellule hôte. L'expression du transgène entraîne une libération du peptide marqueur dans le fluide corporel extracellulaire du mammifère proportionnellement à la quantité du produit de transgène. Le taux de peptide marqueur libéré sert d'indicateur du taux d'expression de transgène. L'expression du transgène confère l'activité du symporteur iodure de sodium (NIS) à une cellule hôte qui exprime le transgène. L'administration ultérieure de iodure marqué entraîne le transport du iodure marqué dans la cellule portant le NIS, qui peut ensuite être localisé et mesuré à l'aide de techniques d'imagerie standard. Le système est particulièrement utile pour surveiller l'expression et la localisation de transgènes thérapeutiques et pour la production du produit génique thérapeutique.
