The invention disclosed a total synthesis process of novel ethoxy combretastatins and their prodrugs. Combretastatins are chemically modified by ethoxy substituted on the 4'-position of their B aryl ring and are converted to be their soluble prodrugs of phosphates or their inner salts of phosphorylcholine by modifying the hydroxyl on the 3'-position of their B aryl ring. Similarly, 3'-amino combretastatin is 4'-ethoxy chemically modified and further side chain of amino acid can be introduced to the amino to form soluble prodrug of amino acidamide. The structure of the said compounds is showed as formula (1). Ethoxy combretastatins possess potent tubulin polymerizaion inhibitory activity and can be used for the treatment of inhibiting tumor or neovascular.
Cette invention concerne un procédé de synthèse totale de nouvelles éthoxy-combretastatines et de leurs promédicaments. Les combretastatines sont modifiées chimiquement par éthoxy substitué à la position 4' de leur noyau B-aryle et converties en leurs promédicaments solubles de phosphates ou leurs sels internes de phosphorylcholine par modification de l'hydroxyle à la position 3' de leur noyau B aryle. De même, la 3'-amino-combretastatine est modifiée chimiquement par 4' éthoxy, et une autre chaîne latérale d'aminoacide peut être introduite dans l'amino pour former un promédicament soluble d'amino-acidamide. La structure desdits composés est indiquée comme formule (1). Les éthoxy-combretastatines présentent une activité puissante inhibitrice de la polymérisation de la tubuline et peuvent être utilisées pour un traitement destiné à inhiber des tumeurs ou un traitement d'états néovasculaires.
