The present invention relates to a method for quickly determining cAMP content or an adenylate cyclase activity in a biological sample containing non-cyclic adenine nucleotides without the use of radioactive agents. Particularly, the present invention provides a method of determining cAMP content or an adenylate cyclase activity in a biological sample containing non-cyclic adenine nucleotides selected from the group consisting of cAMP produced by endogenous adenylate cyclase, and AMP, ATP, ADP and a mixture thereof, which comprises (1) combining a biological sample with effective amounts of apyrase, adenosine deaminase and alkaline phosphatase to enzymatically remove non-cyclic adenine nucleotides other than cAMP, and glucose-6-phosphate in the sample; (2) enzymatically converting cAMP into AMP; (3) determining an amount of AMP without the use of radioactive agents, and a kit to carry out the method.

L'invention concerne une méthode de dosage rapide du contenu cAMP ou de l'activité de l'adénylate cyclase dans un échantillon biologique contenant des nucléotides adénines endogènes non cycliques, et ce sans avoir recours à des réactifs radioactifs. Cette invention concerne plus particulièrement une méthode de dosage du contenu cAMP ou de l'activité de l'adénylate cyclase dans un échantillon biologique contenant des nucléotides adénines non cycliques choisis dans le groupe composé de cAMP formé par une adénylate cyclase endogène, ATP, AMP, ADP, et leurs mélanges. Cette méthode consiste: 1) à ajouter des quantités efficaces d'apyrase, d'adénosine déaminase, et d'alcaline phosphatase à l'échantillon susmentionné, afin de permettre une élimination enzymatique des nucléotides adénines non cycliques autres que cAMP, et du gyclose-6-phosphate provenant de ceux-ci; 2) à effectuer une conversion enzymatique de cAMP en AMP; et 3) à quantifier AMP sans avoir recours à une quelconque substance radioactive. Cette invention concerne enfin un kit permettant d'appliquer cette méthode.
