Polynucleotides encoding carcinoembryonic antigen (CEA) fusion proteins are provided, the CEA fusion proteins comprising a CEA protein, or functional variant thereof, fused to a substantial portion of an immunoenhancing element. The polynucleotides of the present invention can elicit an immune response in a mammal, which, in preferred embodiments, is stronger than the immune response elicited by a wild-type CEA. The gene encoding CEA is commonly associated with the development of human carcinomas. The present invention provides compositions and methods to elicit or enhance immunity to the protein product expressed by the CEA tumor-associated antigen, wherein aberrant CEA expression is associated with a carcinoma or its development. This invention specifically provides adenoviral vector and plasmid constructs carrying polynucleotides encoding CEA fusion proteins and discloses their use in vaccines and pharmaceutical compositions for preventing and treating cancer.
La présente invention se rapporte à des polynucléotides codant des protéines de fusion à antigène carcinoembryonique (CEA), lesdites protéines de fusion CEA comportant une protéine CEA, ou un variant fonctionnel de cette protéine, fusionnée à une partie importante d'un élément immunostimulateur. Les polynucléotides de la présente invention peuvent déclencher une réponse immunitaire chez un mammifère, ladite réponse immunitaire étant, dans des modes de réalisation préférés, plus forte que la réponse immunitaire déclenchée par un CEA du type sauvage. Le gène codant CEA est associé généralement au développement des carcinomes humains. La présente invention se rapporte à des compositions et à des méthodes de déclenchement ou de stimulation de l'immunité au produit protéique exprimé par l'antigène spécifique des tumeurs CEA, une expression de CEA aberrante étant associée à un carcinome ou à son développement. Cette invention se rapporte de manière spécifique à un vecteur adénoviral et à des constructions plasmidiques portant des nucléotides codant des protéines de fusion CEA, ainsi qu'à leur utilisation dans des vaccins et compositions pharmaceutiques pour la prévention et le traitement du cancer.
