An apparatus, product formulation, and method for improved dermal permeation of pharmaceuticals wherein the apparatus includes a thin drug formulation reservoir and a heat-generating chamber separated by a first non-permeable wall, wherein the reservoir and chamber are formed in or supported by a housing. The drug formulation reservoir houses or is capable of housing a predetermined amount of a formulation containing pharmaceutically-active agent(s). The heat- generating/temperature-regulating chamber includes a medium for generating controlled heat, preferably a chemical composition made of carbon, iron, water and/or salt which is activated upon contact with air (oxygen). The function of the heat-generating/temperature-regulating element is to heat the user's skin, rapidly bring the skin temperature to a desired and elevated narrow range and keep it in this range for sufficient time to obtain more rapid, enhanced and less variable dermal absorption of selected pharmaceutically-active agents and to obtain improved clinical effets. Structure for controlling the generation of heat is also disclosed. The apparatus may optionally include a spacing or standoff structure which spans the drug formulation reservoir between the non-permeable wall and the user's skin surface for maintaining a predetermined thickness of the drug formulation on the user's skin surface. Also, a novel product formulation which can be used with the apparatus which uses high percentage of eutectic mixture of local anesthetics to reduce the overall degradation rate of the local anesthetic compound(s) in formulations which are subject to hydrolysis.
La présente invention concerne un appareil, une formulation de produit et un procédé améliorant la perméation cutanée de produits pharmaceutiques. L'appareil comporte un réservoir mince pour formulation médicamenteuse et une chambre thermogénératrice séparée par une première paroi imperméable, le réservoir et la chambre étant formés ou contenus dans un logement. Le réservoir pour formulation médicamenteuse renferme ou est capable de renfermer une dose prédéfinie d'une formulation contenant un ou plusieurs agents pharmaceutiquement actifs. La chambre thermogénératrice/thermorégulatrice renferme un milieu de génération régulée de chaleur, de préférence une composition chimique faite de carbone, de fer, d'eau et/ou de sel qui s'active au contact de l'air (l'oxygène). La fonction de l'élément thermogénérateur/thermorégulateur est de chauffer la peau de l'utilisateur, d'amener rapidement la température de la peau de l'utilisateur dans une plage étroite et élevée, et de la maintenir dans cette plage pendant assez longtemps pour obtenir une absorption cutanée plus rapide, améliorée et moins aléatoire des agents pharmaceutiquement actifs sélectionnés, et d'obtenir un meilleur rendement clinique. L'invention concerne également une structure de régulation de la génération de chaleur. L'appareil peut éventuellement comporter un dispositif d'écartement et d'éloignement qui, afin de conserver une épaisseur prédéterminée de formulation médicamenteuse sur la surface cutanée de l'utilisateur, couvre le réservoir à formulation médicamenteuse entre la paroi imperméable et la surface cutanée de l'utilisateur. L'invention concerne enfin une nouvelle formulation de produit utilisable avec cet appareil et qui nécessite un pourcentage élevé de mélange eutectique d'anesthésique local, ce qui, dans les formulations soumises à hydrolyse, permet de réduire la vitesse de dégradation globale de composé(s) anesthésique(s) local (locaux).
