A process for the enantioselective enzymatic reduction of compounds which comprise a steroid structure (ABCD) including one or several heteroatoms, one or several double bonds and/or one aromatic component in the ring structure and have at least one oxo group at position 3, 7, 11, 12 or 17 in the steroid ring system or in the .alpha.- position at any carbon moiety on the steroid skeleton (=oxosteroid compound), (see formula above) wherein the oxosteroid compound is reduced with a hydroxysteroid dehydrogenase in the presence of a cofactor NADH or NADPH, is characterized in that a) the oxosteroid compound is provided in the reaction at a concentration of >= 50 g/l, b) the oxidized cofactor NAD or NADP formed by the hydroxysteroid dehydrogenase is regenerated continuously by oxidation of a secondary alcohol of general formula R x R y CHOH, wherein R x, R y independently represent hydrogen, a branched or unbranched C1-C8-alkyl and C total >= 3, or by oxidation of a C4-C6-cycloalkanol, and c) an additional oxidoreductase/alcohol dehydrogenase is used for the oxidation of the secondary alcohol of general formula R x R y CHOH or of the cycloalkanol, respectively.
L'invention concerne un procédé de réduction enzymatique énantio-sélective de composés qui comprennent une structure stéroïde (ABCD), y compris un ou plusieurs hétéroatomes, une ou plusieurs doubles liaisons et/ou un groupe aromatique dans la structure cyclique, et qui présentent au moins un groupe oxo en position 3, 7, 11, 12 ou 17 du système cyclique stéroïde ou en position a sur un résidu carbone quelconque dans l'ossature stéroïde (= composé oxostéroïde/composé hydroxystéroïde), le composé oxostéroïde étant réduit avec une hydroxystéroïde déshydrogénase en présence d'un cofacteur NADH ou NADPH, caractérisé en ce que a) le composé oxostéroïde est présent à une concentration > 50 g/l dans le mélange réactionnel, b) le cofacteur NAD ou NADP oxydé formé par l'hydroxystéroïde déshydrogénase est régénéré en continu par oxydation d'un alcool secondaire de formule générale R<SUB>x</SUB>R<SUB>y</SUB>CHOH, où R<SUB>x</SUB>, R<SUB>y</SUB> sont indépendamment l'un de l'autre un hydrogène ou un alkyle en C1 à C8 ramifié ou non ramifié et C<SUB>total</SUB> = 3, ou bien par oxydation d'un cycloalcanol en C4 à C6, et c) une oxydoréductase/alcool déshydrogénase supplémentaire est ajoutée pour l'oxydation de l'alcool secondaire de formule générale R<SUB>X</SUB>R<SUB>Y</SUB>CHOH ou du cycloalcanol.
