The present invention relates to methods for detecting free floating nucleic acids, as present in not cellular bound nucleic acids in bodily fluids like plasma or serum fractions of human or animal blood or in any other tissue samples derived from the human or animal body in order to diagnose a cell proliferative disease. Specifically the invention relates to the detection of increased levels of nucleic acids in bodily fluids. Furthermore the invention allows to determine the source of the enriched DNA by measuring the ratio of DNA originating from a certain organ versus total DNA from other organs in a given bodily fluid sample by specifying the DNA's methylation pattern. This can be done with or without increasing the DNA concentration of a given biological sample. In a preferred embodiment a further analysis of this methylation pattern allows for the detection of the presence of tumourous or otherwise proliferative disease in said organ.
La présente invention concerne des méthodes permettant de détecter des acides nucléiques flottant librement, tels qu'ils apparaissent dans des acides nucléiques qui ne sont pas liés à des cellules, dans des liquides organiques, tels que les fractions plasmatiques ou sériques du sang humain ou animal, ou dans n'importe quel autre échantillon tissulaire prélevé sur un corps humain ou animal, pour diagnostiquer une pathologie faisant intervenir une prolifération cellulaire. L'invention concerne, plus particulièrement, la détection de taux élevés d'acides nucléiques dans les liquides organiques. Par ailleurs, l'invention concerne également une méthode permettant de déterminer la source de l'ADN enrichi, consistant à mesurer le taux d'ADN provenant d'un organe donné par rapport à l'ADN total provenant d'autres organes, dans un échantillon de liquide organique donné, par spécification du profil de méthylation de l'ADN. Cette étape peut être réalisée avec ou sans augmentation de la concentration d'ADN d'un échantillon biologique donné. Dans un mode de réalisation préféré, la méthode consiste à approfondir l'analyse de ce profil de méthylation pour détecter la présence de pathologies tumorales ou de pathologies liées à une prolifération cellulaire, dans l'organe étudié.
