The present invention relates to a method for the preparation of citalopram comprising reaction a compound of formula (I) with a compound having formula (II) wherein X is a suitable leaving group and R is -CH2-O-Pg, -CH2-NPg1Pg2 , - CO-N(CH3)2, -CH(OR1)(OR2), -C(OR4)(OR5)(OR6), -COOR3, -CH2-CO-NH2, -CH=CHR7 and -CONHR8 wherein Pg is a protection group for an alcohol group, Pg1 and Pg2 are protection groups for an amino group, R1 and R2 are independently selected from alkyl, alkenyl, alkynyl and optionally alkyl substituted aryl or aralkyl groups or R1 and R2 together form a chain of 2 to 4 carbon atoms, each of R3, R4, R5, R6 and R7 are independently selected from alkyl, alkenyl, alkynyl and optionally alkyl substituted aryl or aralkyl and R8 is hydrogen or methyl; to form a compound of formula (III) wherein R is as defined above; followed by conversion of the group R to form a dimethylaminomethyl group and isolation of citalopram base or a pharmaceutically acceptable salt thereof.
La présente invention concerne un procédé convenant à l'élaboration de citalopram et consistant à prendre un composé représenté par la formule (I) et à le faire réagir avec un composé représenté par la formule (II). Dans ces formules, X est un groupe quittant. R est -CH¿2?-O-Pg, -CH¿2?-NPg¿1?Pg¿2?, -CO-N(CH¿3?)¿2?, -CH(OR?1¿)(OR?2¿), -C(OR?4¿)(OR?5¿)(OR?6¿), -COOR?3¿, -CH¿2?-CO-NH¿2?, -CH=CHR?7¿ ou -CONHR?8¿. Pg est un groupe de protection destiné à un groupe alcool. Pg¿1? et Pg¿2? sont des groupes de protection destinés à un groupe amino. R?1¿ et R?2¿ appartiennent indépendamment au groupe des alkyle, alcényle, alkynyle et aryle ou aralkyle éventuellement substitués par alkyle. R?1¿ et R?2¿ peuvent former ensemble une chaîne de 2 à 4 atomes de carbone. Chacun des R'3¿, R?4¿, R?5¿, R?6¿ et R?7¿ appartiennent indépendamment au groupe des alkyle, alcényle, alkynyle et aryle ou aralkyle éventuellement substitués par alkyle. R?8¿ est hydrogène ou méthyle. Cette réaction se fait de façon à aboutir à un composé représenté par la formule (III) dans laquelle R est tel que défini précédemment. Cette réaction est alors suivie d'une conversion du groupe R de façon à former un groupe diméthylaminométhyle, puis de l'isolement de la base citalopram ou de l'un de ses sels pharmaceutiquement admis.
