The invention is directed to a method for treating both the early and late phases of allergic asthma by introducing naked polynucleotides which operatively encode for the asthma-initiating antigen into the host. The antigen-encoding polynucleotides are administered to host tissues which contain a high concentration of antigen presenting cells (e.g., skin and mucosa) relative to other host tissues. Expression of the asthma-initiating antigen encoding polynucleotides of the invention inside of antigen presenting cells (without substantial secretion therefrom) induces antigen tolerance while suppressing IgE antibody formation in the early phase of the disease, and also suppresses cytokine-mediated eosinophil accumulation in the late phase of the disease. Devices and compositions for use in the methods of the invention are also described.
L'invention se rapporte à une méthode de traitement de la phase initiale et de la phase finale d'asthme allergique, qui consiste à introduire dans l'hôte des polynucléotides nus, lesquels codent de façon opérationnelle l'antigène déclenchant l'asthme. Les polynucléotides codant l'antigène sont administrés à des tissus hôtes qui contiennent une forte concentration de cellules présentant l'antigène (par exemple, de la peau ou des muqueuses), par rapport à d'autres tissus hôtes. L'invention a pour objet l'expression desdits polynucléotides codant l'antigène déclenchant l'asthme dans des cellules présentant l'antigène (sans sécrétion importante de celles-ci), expression qui induit une tolérance à l'antigène tout en supprimant la formation d'anticorps IgE dans la phase initiale de la maladie et l'accumulation d'éosinophile véhiculé par la cytokine dans la phase finale de la maladie. L'invention se rapporte également à des dispositifs et des compositions destinés à être utilisés dans les méthodes décrites dans l'invention.
