The proposed process for the qualitative/semi-quantative détermination of progesterone in a biological fluid on an ELISA principle comprises the following steps. A few drops of the fluid to be examined are taken, they are diluted with an aqueous sample diluent solution, this mixture is put into contact with a surface coated with a specific antibody and a progesterone-enzyme conjugate solution is added which preferably reacts with still free bond positions of the antibodies which are fixed on a surface. Incubation mixture and unbonded reaction partners are removed by being washed off the surface and the bonded enzyme is detected by colour reaction. The colour developed is a measure of the progesterone content of the fluid to be examined. The process of the invention is particularly suitable in a fast test for the simple and reliable detection of the insemination and conception time and monitoring the pregnancy of domestic animals. Thereby it is possible for the first time very reliably to distinguish between the oestrus and pregnancy period and intermediate the period of low progesterone concentrations and largely avoid the many faulty diagnoses and interpretations commonly made with prior art processes.
