The present invention provides canine heavy chain and light chain (both .lambda. and .kappa.) variable domain nucleic acids and polypeptides. The present invention also provides caninized antibodies comprised of the present disclosed canine variable domain framework sequences and CDRs specific for an antigen obtained from antibodies from other than canines (preferably mouse). The invention also provides methods of making caninized antibodies and using them. This disclosure specifically describes caninized antibodies that are useful in the treatment of canine allergy. The general process of caninization permits the development of useful therapeutics, which includes but is not limited to allergy, osteoarcomma's and lymphomma's.

L'invention concerne des polypeptides et des acides nucléiques canins à domaine variable de chaîne lourde et de chaîne légère (à la fois .lambda. et .kappa.). L'invention concerne également des anticorps canins contenus dans des séquences d'infrastructure de domaine variable canin de la présente invention et spécifiques à la CDR pour un antigène obtenu à partir d'anticorps autres que celui provenant de canins (de préférence de souris). L'invention concerne également des procédés de fabrication d'anticorps pour canins et leur utilisation. La description décrit de manière spécifique des anticorps pour canins qui sont utiles dans le traitement d'allergie canine.
