The invention concerns a method for rapidly quantifying living coliform microorganisms in a water sample which consists in contacting the microorganisms with a fluorogenic substrate which is hydrolyzed in methylumbelliferone by an enzyme released by the microorganisms. The methylumbelliferone is detected by the fluorescence it emits, the speed of variation of said fluorescence being measured so as to determine the concentration of coliform microorganisms in the original sample based on the calculated speed of variation and on a correlation scheme. Typically, the method consists in carrying out a preliminary step of treating, in particular by pre-filtering, the sample to remove at least part of the particles suspended in the water, thereby allowing a quantification quite comparable to that of the method normalized by microplate. The invention is applicable to swimming water for detecting Escherichia Coli.
L'invention concerne un procédé de quantification rapide des micro-organismes coliformes vivants dans un échantillon d'eau dans lequel on place les micro- organismes en contact avec un substrat fluorogène qui est hydrolyse en methylumbelliferone par une enzyme dégagée par les micro-organismes. La methylumbelliferone est détectée par la fluorescence qu'elle émet, la vitesse de variation de cette fluorescence étant mesurée afin de déterminer la concentration des micro-organismes coliformes dans l'échantillon d'origine à partir de la vitesse de variation calculée et d'un schéma de corrélation. De façon caractéristique, on réalise une étape préliminaire de traitement, notamment une pré-filtration, de l'échantillon permettant de retirer au moins une partie des particules en suspension dans l'eau, ce qui permet de donner une quantification tout à fait comparable à celle de la méthode normalisée par microplaque. Application à l'analyse des eaux de baignade pour la détection des Escherichia Coli.
