The present invention relates to a method of amplifying a nucleic acid sequence present in a first strand of a double stranded nucleic acid molecule wherein said molecule incorporates an unmodified recognition site for a restriction enzyme capable of cutting the first strand at the 5' end of the sequence therein to be amplified and leaving the 3'-end region of the second strand projecting beyond the cut site in the first strand. The method further comprises treating said molecule with said enzyme in the presence of a strand displacing polymerase and unmodified nucleotides for incorporation in an extending nucleic acid strand such that there is or becomes hybridised to said 3'-end region of the second strand a primer sequence complementary thereto whereby said primer sequence is extended in the 5' to 3' direction using the second strand as a template to regenerate the restriction endonuclease cut site and displace the sequence to be amplified.

L'invention concerne un procédé d'amplification d'une séquence d'acide nucléique présente dans un premier brin d'une molécule d'acide nucléique à double brin, ladite molécule incorporant un site de reconnaissance non modifié pour une enzyme de restriction capable de couper le premier brin à l'extrémité 5' de la séquence à amplifier et de laisser la région terminale 3' du second brin, en saillie au-delà du site de coupure dans le premier brin. Le procédé est caractérisé en ce qu'on traite ladite molécule avec ladite enzyme en présence d'une polymérase déplaçant le brin et de nucléotides non modifiés pour l'incorporation dans un brin d'acide nucléique en extension, dans des conditions telles qu'il soit ou devienne hybridisé en ladite région terminale 3' du second brin d'une séquence primaire complémentaire, de telle sorte que ladite séquence primaire soit étendue dans la direction 5' au 3' en utilisant le second brin comme modèle pour régénérer le site de coupure d'endonucléase de restriction et déplacer la séquence à amplifier.
