A method of preparing autologus T-lymphocytes for re-introduction into a patient having cancer, which method comprises obtaining peripheral blood mononuclear cells (PBMCs) from a patient immunized with an antigen of the canncer, stimulating the PBMCs with the antigen of the cancer in vitro, transducing the PBMCs with a retrovial vector, which (a) comprises and expresses a human interleukin-2 (IL-2) coding sequence operably linked to a retroviral promoter, (b) does not comprise an exogenously introduced gene that enables phenotypic selection, and (c) comprises a viral envelope that efficiently transduces CD8+ T-lymphocytes; a method of preparing autologous tumor-infiltrating lymphocytes (TILs) for reintroduction into a patient having cancer, which method comprises obtaining TILs from a tumor of the patient, optionally immunized with an antigen of the cancer, optionally stimulating the TILs with the antigen of the cancer in vitro, and transducing the TILs with the aforementioned retroviral vector; compositions comprising cells obtained in accordance with such methods; and methods of treating a patient having cancer by administering to the patient cells obtained in accordance with such methods or compositions comprising same.
La présente invention concerne un procédé permettant d'obtenir des lymphocytes T autologues à réintroduire chez un patient cancéreux. A cet effet, chez un patient immunisé avec un antigène du cancer, on prélève des cellules monunucléées de sang périphérique ou PBMC (Peripheral Blood Mononuclear Cells), on stimule in vitro les PBMC avec l'antigène du cancer, et en transduit avec un vecteur rétroviral les PBMC. Ce vecteur doit (a) comprendre et exprimer, fonctionnellement liée à un promoteur rétroviral une séquence codant l'interleukine 2 (IL-2) humaine, (b) ne pas comprendre de gène introduit de façon exogène qui permette une sélection phénotypique, et (c) comprendre une enveloppe virale qui transduise de façon efficace les lymphocytes T CD8+. L'invention concerne également un procédé permettant d'obtenir des lymphocytes infiltrant les tumeurs ou TIL (Tumor-Infiltrating Lymphocytes) à réintroduire chez un patient cancéreux. A cet effet, chez un patient, éventuellement immunisé avec l'antigène du cancer, on prélève des TIL d'une tumeur, on stimule éventuellement in vitro les TILS avec l'antigène du cancer, puis on transduit les TIL avec le vecteur rétroviral déjà mentionné. L'invention concerne aussi des compositions comprenant des cellules obtenues selon de tels procédés. L'invention concerne enfin des procédés concourant au traitement d'un patient cancéreux par administration à ce patient de cellules obtenues au moyen de tels procédés ou de compositions les comprenant.
