Abstract Substrates for .beta.-lactamase of general formula (I) in which one of X and Y is a fluorescent donor moiety and the other is a quencher (which may or may not re-emit); R' is selected from the group consisting of H, lower (i.e., alkyl of 1 to about carbon atoms) and (CH2)n OH, in which n is 0 or an integer from 1 to 5; R" is selected from the group consisting of H, physiologically acceptable metal and ammonium cations, -CHR2OCO(CH2)n CH3, - CHR2OCOC(CH3)3, acylthiomethyl, acyloxy- alpha-benzyl, delta-butyrolactonyl, methoxycarbonyloxymethyl, phenyl, methylsulphinylmethyl, beta-morpholinoethyi, dialkylaminoethyl, acyloxyalkyl, dialkylaminocarbonyloxymethyl and aliphatic, in which R2 is selected from the group consisting of H and lower alkyl; A is selected from the group consisting of S, O, SO, SO2 and CH2; and Z' and Z" are linkers for the fluorescent donor and quencher moieties. Methods of assaying .beta.-lactamase activity and monitoring expression in systems using .beta.-lactamase as a reporter gene also are disclosed.

Cette invention concerne des substrats pour beta -lactamase représentés par la formule générale (I) dans laquelle si X est une fraction de donneur fluorescent, Y est un extincteur de luminescence (qui peut ou non ré-émettre) ou vice versa, R' appartient au groupe constitué de H, d'un alkyle inférieur (c'est-à-dire alkyle comportant de 1 à 5 atomes de carbone) et de (CH2)nOH, où n est 0 ou un entier compris entre 1 et 5, R'' appartient au groupe constitué de H, de cations métalliques et ammonium physiologiquement acceptables, de -CHR<2>OCO(CH2)nCH3, - CHR<2>OCOC(CH3)3, acylthiométhyle, acyloxy-alphabenzyle, delta-butyrolactonyle, méthoxycarbonyloxyméthyle, phényle, méthylsulphinylméthyle, béta-morpholinoéthyle, dialkylaminoéthyle, acyloxyalkyle, dialkylaminocarbonyloxyméthyle et d'un composé aliphatique, où R<2> est sélectionné dans groupe constitué de H et d'un alkyle inférieur, A est sélectionné dans le groupe constitué de S, O, SO, SO2 et CH2, et Z' et Z'' sont des agents de liaison pour les fractions de donneur fluorescent et d'extincteur de luminescence.
