The invention provides a method of using chip-based gel electrophoresis to determine the presence of polypeptides with selected disulfide linkage patterns, for example, completely formed tetrameric antibodies as compared to incompletely formed heterodimeric half-antibodies. The invention further features a kit, comprising a chip, and instructions for conducting the foregoing method. The methods and kits of the invention are amendable to high throughput applications for the monitoring of production and quality control of recombinant therapeutic antibodies.
La présente invention concerne un procédé pour utiliser une électrophorèse sur gel sur circuits intégrés, afin de déterminer la présence de polypeptides à motifs de liaison disulfure choisis, par exemple des anticorps tétramères complètement formés, par comparaison à des demi-anticorps hétérodimères incomplètement formés. La présente invention concerne également une trousse comprenant un circuit intégré et des instructions pour mettre en oeuvre ledit procédé. Les procédés et les trousses selon cette invention peuvent être modifiés pour des applications à haut rendement afin de contrôler la production et la qualité d'anticorps thérapeutiques de recombinaison.
