The detection of optical stripes on an image of a glass slide (20) of material of interest. An automated microscope system (511) with an illuminator (508) having a spatial intensity homogenizer (40) provides spatially uniform illumination of a microscope slide specimen (20). Periodic optical patterns formed from the illumination system (508) are detected by a computer system (540). The computer (540) forms a high transmission mask (717) of an image (712) of the biological specimen (20). Edges in the image are detected (716) and a low transmission area process (718) is performed. Horizontal stripes are enhanced (720). A final thresholding (722) generates a binary image (727) of the patterns. Stripes in the patterns are detected (724) and a stripe flag (706) and stripe area (708) are calculated.
