The present Invention relates to novel fusidic acid derivatives of general formula (I) wherein X represents halogen, trifluoromethyl, cyano, azido, alkyl, alkenyl or aryl, wherein. said alkyl, alkenyl or aryl are optionally substituted by one or more, same or. different substituents selected from the group consisting of alkyl, alkenyl, aryl, alkoxy, nitro, alkylthio, halogen, azido, trifluoromethyl and cyano; Y and Z both' represent hydrogen, or together with the C-17/C-20 bond form a double bond between C-17 and C-20, or together are methylene and form a cyclopropane ring in combination with C-17 and C-20; A represents a bond,. O, S or S(O); B represents C1-6 alkyl, C2-6 alkenyl, CI-6 acyl, C3-7cycloalkylcarbonyl or benzoyl, all of which are optionally substituted with one or more substituents selected from the group consisting of halogen, hydroxy, alkoxy, aryl, heteroaryl and azido, or, if A represents a bond, B may also represent hydrogen; Q1 and 'Q2 independently represent. -CH2-, -C(O)-, -(CHOR)-, -(CHOR)-, -(CHSH)-, - (NH)-, -(CHNH2)- or - (CHW)-., wherein R represents CI-6alkyl and W represents halogen, cyano, azido or trifluoromethyi; Q3 represents -CH2-, -C(O)- or -CHOH-; G represents hydrogen, OH or O-CO-CH3; two bonds in the pentacyclic ring being depicted with full and dotted lines to indicate that either of the two bonds may be a double bond, in which case Y is absent and Z represents hydrogen; the bond between C-1 and C-2 being either a single or a double bond; and pharmaceutically acceptable salts and easily hydrolysable esters thereof, to pharmaceutical compositions comprising said derivatives, as well as to their use in therapy.
L'invention concerne des nouveaux dérivés d'acide fusidique de formule générale (I) dans laquelle X représente halogène, trifluorométhyle, cyano, azido, alkyle, alcényle ou aryle ; lesdits alkyle, alcényle ou aryle étant éventuellement substitués par un ou plusieurs substituants identiques ou différents sélectionnés dans le groupe comprenant alkyle, alcényle, aryle, alcoxy, nitro, alkylthio, halogène, azido, trifluorométhyle et cyano ; Y et Z représentent tous deux hydrogène, ou forment avec la liaison C-17/C-20 une double liaison entre C-17 et C-20, ou représentent ensemble méthylène et forment un anneau cyclopropane en combinaison avec C-17 et C-20 ; A représente une liaison, O, S ou S(O) ; B représente C¿1?-¿6 ?alkyle, C2-¿6? alcényle, C<SB>I</SB>-6 acyle, C¿3?-¿7 ?cycloalkylcarbonyle ou benzoyle, tous étant éventuellement substitués par un ou plusieurs substituants sélectionnés dans le groupe comprenant halogène, hydroxy, alcoxy, aryle, hétéroaryle et azido, ou, si A représente une liaison, B peut également représenter hydrogène ; Q1 et Q2 représentent indépendamment -CH2-, -C(O)-, -(CHOR)-, -(CHOR)-, -(CHSH)-, - (NH)?-¿, -(CHNH¿2?)- ou -(CHW)-, R représentant C<SB>I</SB>-¿6?alkyle et W représentant halogène, cyano, azido ou trifluorométhyle ; Q3 représente -CH2-, -C(O)- ou -CHOH- ; G représente hydrogène, OH ou O-CO-CH¿3? ; deux liaisons dans l'anneau pentacyclique étant décrites avec des lignes pleines et en pointillés afin d'indiquer que l'une ou l'autre des deux liaisons peut être une double liaison, auquel cas Y est absent et Z représente hydrogène ; la liaison entre C-1 et C-2 étant une liaison simple ou double ; et des sels acceptables sur le plan pharmaceutique et des esters facilement hydrolysables desdits dérivés, des compositions pharmaceutiques comprenant ces derniers, ainsi que leur utilisation thérapeutique.
