A DNA vaccine effective for eliciting an immune response against cell that present a carcinoembryonic antigen (CEA) comprises a DNA operably encoding a CEA and a DNA operably encoding a CD40 ligand, SEQ ID NO:1 and SEQ ID NO:2, respectively, or its homotrimer, CD40LT. The DNA vaccine can be incorporated in a delivery vector such as an attenuated live bacterium or virus, or a liposome carrier. In a method embodiment, the DNA vaccine is administered orally to a mammal, such as a human, to elicit an immune response against CEA presenting cells such as colon cancer cells. A preferred method embodiment includes the additional step of treating the mammal with recombinant antibody fusion protein huKS1/4-IL2 to enhance the immune response effectiveness of the vaccine.
La présente invention concerne un vaccin à base d'ADN permettant de déclencher une réponse immunitaire dirigée contre une cellule à antigène carcino-embryonnaire (ACE). Ce vaccin comporte, d'une part un ADN codant fonctionnellement un ACE (SEQ ID NO:1) et un ADN codant fonctionnellement un ligand CD40 (SEQ ID NO:2), ou son homotrimère CD40LT. Ce vaccin à base d'ADN peut s'intégrer, soit à un vecteur tel qu'une bactérie ou un virus vivant atténué, soit à un liposome transporteur. Selon un mode de réalisation du procédé, on administre par voie orale le vaccin à base d'ADN à un mammifère, tel qu'un humain, pour déclencher une réponse immunitaire contre un des cellules portant un ACE, notamment, des cellules du cancer du colon. Un mode de réalisation préféré du procédé comporte une étape supplémentaire, en l'occurrence, le traitement du mammifère au moyen d'une protéine de fusion d'anticorps recombinant huKS1/4-IL2 pour obtenir une réponse immunitaire plus puissante du vaccin.
