Methods are provided for the rapid isolation of highly purified and functionally intact dendritic cells from a mixed cell population, using colloidal superparamagnetic particles. Dendritic cells are enriched from a blood or lymph sample using a two step high-gradient magnetic cell separation. Lymphocytes, natural killer and monocytic cells are depleted by specific binding to markers present on lymphoid and myeloid cells. In a separate step, dendritic cells are enriched by HGMS. Purified dendritic cells are useful as a source of antigen presenting cells for in vitro analysis, and in immunomodulating therapies, particularly for priming naive T cells.

L'invention concerne des procédés d'isolement rapide de cellules dendritiques hautement purifiées et fonctionnellement intactes à partir d'une population de cellules mélangées, à l'aide de particules colloïdales superparamagnétiques. Les cellules dendritiques sont enrichies à partir d'un échantillon de sang ou de lymphe par une séparation magnétique de cellules à gradient élevé en deux étapes. Les lymphocytes, les cellules tueuses naturelles et monocytaires sont épuisées par liaison spécifique à des marqueurs présents sur des cellules lymphoïdes et myéloïdes. Dans une étape séparée, des cellules dendritiques sont enrichies par HGMS (triage magnétique à gradient élevé). Les cellules dendritiques purifiées sont utiles en tant que source de cellules de présentation d'antigène pour une analyse in vitro, et dans des thérapies d'immunomodulation, notamment pour amorcer des lymphocytes T naïfs.
