This invention discloses a novel method for conjugating RNA oligonucleotides with other molecular entities exclusively at the 5'-position of the oligonucleotide. Specifically, this invention discloses a method for conjugating or derivatizing RNA oligonucleotides via transcription in the presence of a 5'-modified guanosine. Included in the invention are the novel bioconjugated oligonucleotides that can be prepared according to the method of the invention.
