The present invention is directed to a method for delivering exogenous proteins to the cytosol, by binding a target antigen (such as a protein) to a transport factor that contains a fragment of a bipartite protein exotoxin, but not the corresponding protective antigen. Preferably, the target antigen is fused to the transport factor. Preferred transport factors include the protective antigen binding domain of lethal factor (LFn) from B.anthracis, consisting of amino acids 1-255, preferably a fragment of at least 80 amino acids that shows at least 80 % homology to LFn, and a fragment of about 105 amino acids from the carboxy portion that does not bind PA. The target antigen can include any molecule for which it would be desirable to elicit a CMI response, including viral antigens and tumor antigens.
L'invention concerne une méthode de distribution de protéines exogènes au cytosol, par liaison d'un antigène cible (tel qu'une protéine) à un facteur de transport contenant un fragment d'exotoxine protéique bipartite, ledit antigène ne se pas à l'antigène protecteur correspondant. Ledit antigène cible fusionne, de préférence, avec le facteur de transport. Les facteurs de transport préférés comprennent le domaine de liaison de l'antigène protecteur du facteur létal (LFn) provenant de <i>B.anthracis</i>, constitué d'acides aminés 1-255, de préférence un fragment d'au moins 80 acides aminés présentant au moins 80 % d'homologie avec LFn, et un fragment d'environ 105 acides aminés provenant de la partie carboxy qui ne se lie pas à PA. Cet antigène cible peut comprendre une molécule quelconque pour laquelle il serait souhaitable de déclencher une réponse CMI, notamment des antigènes viraux et tumoraux.
