The invention relates to a process for the preparation of pancreatic carboxypeptidase B or an isoform or a mutein of carboxypeptidase B, where (a) a natural or unnatural precursor form of carboxypeptidase B is expressed in a microorganism by secretion, (b) the precursor form expressed by secretion is purified and (c) the purified precursor form is converted into the active carboxypeptidase B, the isoform or the mutein thereof by an enzymatic treatment, a nucleic acid construct, its use in the process mentioned and its preparation, and a host cell, its preparation and use in a process for the preparation of pancreatic carboxypeptidase B or an isoform or a mutein thereof.

L'invention concerne un procédé de production de carboxypeptidase B pancréatique ou d'une isoforme ou d'une mutéine de carboxypeptidase B. Ce procédé consiste (a) à exprimer par voie sécrétoire une forme de précurseur naturel ou artificiel de carboxypeptidase B dans un micro-organisme, (b) à nettoyer la forme précurseur exprimée par voie sécrétoire et (c) à transformer la forme précurseur nettoyée en carboxypeptidase B active, son isoforme ou sa mutéine par un traitement enzymatique. L'invention concerne également une structure d'acide nucléique, son utilisation dans le procédé mentionné et sa production, ainsi qu'une cellule hôte, sa production et son utilisation dans le procédé de production de carboxypeptidase B pancréatique ou de son isoforme ou de sa mutéine.
