A direct quantitation of RNA contained in a sample is obtained by capillary electrophoresis of the RNA hybridized to a DNA probe of complementary sequence stabilized by the combination of a fluorophore terminally conjugated to the DNA probe and a dye intercalating the RNA-DNA hybrid so formed. The RNA is quantified by measuring the total fluorescence emitted by the electrophoresed hybrid upon excitation by a laser generated light beam. DNA/DNA hybrids may also be detected by this method.
On obtient une quantification directe de l'ARN contenu dans un spécimen par électrophorèse capillaire de l'ARN hybridé à une sonde d'ADN dont la séquence complémentaire est stabilisée par la combinaison d'un fluorophore conjugué en terminaison à la sonde d'ADN et d'un colorant placé entre l'hybride d'ARN et d'ADN obtenu. On quantifie l'ARN par mesure de la totalité de la fluorescence émise par l'hybride soumis à l'électrophorèse et excité par un faisceau de lumière laser. Ce procédé permet également de détecter des hybrides ADN/ADN.
