A method for detecting a de-myelinating disease or spongiform encephalopathy in mammals comprises testing a biological sample obtained from the mammal for IgA antibodies indicative of infection by an Acinetobacter species. The Acinetobacter species is one which presents to the mammal an antigen which exhibits molecular mimicry with the myelin of the mammal e.g. Acinetobacter calcoaceticus. The antibodies tested for are antibodies which bind to an epitope present in or derived from the Acinetobacter species or to a prepared peptide sequence corresponding thereto or to a conformationally similar peptide sequence e.g. the peptide sequence RFSAWGAE or ISRFAWGEV. The method tests for bovine spongiform encephalopathy, multiple sclerosis and Creutzfeldt- Jacob disease in humans. A test kit uses as the test antigen the whole Acinetobacter organism or at least one prepared peptide sequence as described above and a secondary antibody against the human, bovine, or other mammalian IgA.
La présente invention concerne un procédé permettant de détecter chez des mammifères une maladie démyélinisante ou l'encéphalopathie spongiforme. Ce procédé consiste à rechercher dans un échantillon biologique prélevé sur le mammifère des anticorps IgA caractéristiques d'une infection par Acinetobacter. En effet, Acinetobacter est l'une des espèces présentant au mammifère un antigène qui fait preuve d'une aptitude à mimer au niveau moléculaire la myéline du mammifère, comme par exemple Acinetobacter calcoaceticus. Les anticorps recherchés sont des anticorps qui se lient, soit à un épitope qui est présent dans Acinetobacter ou qui en est dérivé, soit encore à une séquence peptidique préparée lui correspondant, soit enfin à une séquence peptidique de conformation semblable telle que la séquence peptidique RFSAWGAE ou ISRFAWGEV. Le procédé permet de détecter l'encéphalopathie spongiforme bovine, et chez l'homme, la sclérose en plaques et la maladie de Creutzfeldt-Jacob. L'invention concerne également un nécessaire de diagnostic utilisant comme antigène test la totalité de l'organisme d'Acinetobacter ou au moins une séquence peptidique préparée telle que décrite précédemment et un anticorps secondaire dirigé contre l'IgA humain, bovin ou d'autres mammifères.
