Novel pentacyclic steroids and pentacyclic D-homosteroids comprising: (i) the tetracyclic steroid ring system or tetracyclic D-homosteroid ring system, respectively; (ii) a C(3) substituent selected from the group consisting of (a) a hydroxyl or carboxyl in the "-configuration and (b) a sulfate or other negatively charged moiety; and (iii) a fused fifth ring, the fused fifth ring comprising a hydrogen bond acceptor, and (a) in the case of the pentacyclic steroid the C(13) and C(17) carbons, or (b) in the case of the pentacyclic D- homosteroid the C(13) and C(17a) carbons, having utility as anesthetics and in the treatment of disorders relating to GABA function and activity.
L'invention concerne de nouveaux stéroïdes et D-homostéroïdes pentacycliques comprenant: i) respectivement, le système cyclique stéroïde tétracyclique ou le système D-homostéroïde tétracyclique; ii) un substituant C(3) sélectionné dans le groupe constitué par a) un hydroxyle ou un carboxyle dans la configuration .alpha., et b) un sulfate ou une autre fraction chargée négativement; et iii) un cinquième cycle fusionné comprenant un accepteur de liaison hydrogène, et a) dans le cas du stéroïde pentacyclique, les carbones C(13) et C(17) ou b) dans le cas du D-homostéroïde pentacyclique, les carbones C(13) et C(17a). Ces nouveaux stéroïdes et D-homostéroïdes pentacycliques sont utilisés comme as anesthésiques et pour traiter des troubles associés à la fonction ou à l'activité GABA.
