Methods for delivering non-mitochondrial proteins to mitochondria are provided. Also provided are nucleic acid constructs comprising a coding sequence encoding a DNA- binding polypeptide, fused to a mitochondrial targeting sequence (MTS) and a nuclear export signal (NES), and the encoded proteins. The construct successfully delivers DNA binding proteins to the mitochondrion. A chimeric methylase based on the above construct is successfully delivered to mitochondria, resulting in modification of mtDNA.

L'invention concerne des procédés de délivrance de protéines non mitochondriales aux mitochondries. Elle concerne également des produits d'assemblage d'acides nucléiques comprenant une séquence codante, qui code pour un polypeptide de liaison à l'ADN, fusionnée à une séquence de ciblage des mitochondries (MTS, mitochondrial targeting sequence) et à un signal d'export nucléaire (NES, nuclear export signal), ainsi que les protéines codées. Le produit d'assemblage permet de délivrer avec succès à la mitochondrie des protéines de liaison à l'ADN. Une méthylase chimérique à base du produit d'assemblage précité est délivrée avec succès aux mitochondries, provoquant une modification de l'ADNmt.
