The present invention relates to use of GPI-PLD antagonists for the prevention, treatment and diagnosis of pre-eclampsia. The substantial GPI-PLD activity is present in the placenta in pre-eclampsia is not expressed in the placenta, but rather is taken up from the maternal circulation. As a result, abnormal or dysregulated GPI-PLD activity present in the placenta in pre- eclampsia may be correctable by administration of GPI-PLD to the mother to correct the problems caused by abnormal or dysregulated GPI-PLD, e.g. to reduce the abnormal release and in situ production of placental IPGs involved in the pathogenesis of pre-eclampsia. This can be achieved using exogenous GPI- PLD or a fragment thereof, e.g. an inactive GPI-PLD capable of competing with or displacing the abnormal or dysregulated GPI-PLD, e.g. from Apo-A1.
L'invention concerne l'utilisation d'antagonistes de phospholipases D spécifiques de glycosyl phosphatidyl inositol (GPI-PLD) pour prévenir, traiter et diagnostiquer une pré-éclampsie. L'activité sensible de GPI-PLD est présente dans le placenta dans la pré-éclampsie et n'est pas exprimée dans le placenta, mais plutôt enlevée de la circulation maternelle. De ce fait, une activité anormale ou déréglée de GPI-PLD présente dans le placenta dans une pré-éclampsie peut être corrigée par une administration de GPI-PLD à la mère, pour corriger les problèmes causés par une activité anormale ou déréglée de GPI-PLD, par exemple réduire la libération anormale ou la production in situ des IPG placentaires impliqués dans la pathogénèse d'une pré-éclampsie. Cette amélioration peut être réalisée par l'utilisation de GPI-PLD exogène ou d'un fragment de celle-ci, par exemple une GPI-PLD inactive capable d'entrer en compétition avec une GPI-PLD anormale ou déréglée ou de la déplacer, par exemple d'Apo-A1.
