The present invention includes an improved method to detect and quantitatively assay the amount of DNA damage in large double-stranded DNA templates starting with less than 50 ng of sample DNA. This invention has utility in vivo for quantitating gene-specific DNA damage, measuring DNA repair rates and monitoring efficacy of anti-neoplasia therapy in patients. Additionally, this invention is useful to detect and assess risk due to mutagenic environmental hazard site, and monitor hazard site dynamics and includes a device for accomplishing these objects.

L'invention porte sur un procédé amélioré de détection et de mesure des quantités d'ADN endommagé dans de grosses matrices d'ADN double brin (à partir d'échantillons de moins de 50 ng d'ADN). L'invention s'applique à la mesure in vivo des dommages causés aux gènes de l'ADN et à celle des taux de réparation de l'ADN, et au suivi de l'efficacité de thérapeutiques antinéoplasiques chez un patient. L'invention s'avère en outre utile pour détecter et évaluer les risques présentés par les sites à risque du fait d'un environnement mutagène et suivre la dynamique des sites à risque. L'invention porte également sur un dispositif destiné à accomplir ces tâches.
