Provided herein is a method for the quantitative screening of hydrolase for desired substrate activity using pH indicators which are sensitive to the release of protons from a chemical reaction in a reaction mixture. The method comprises selecting buffer and indicator conditions such that both have the same affinity for portions such that the relative amount of buffer protonated is proportional to the amount of indicator protonated as the pH of the reaction mixture shifts. A reaction mixture is then prepared comprising a buffer, indicator, hydrolase to be tested, and desired substrate to be tested, allowing the hydrolase to react with the substrate. The reaction is monitored by detection of change in color of the reaction mixture, determined by the affect of the reaction on the indicator.

L'invention concerne une méthode de criblage quantitatif d'une hydrolase, cette méthode permettant de détecter l'activité du substrat souhaitée à l'aide d'indicateurs du pH sensibles à la libération de protons due à une réaction chimique d'un mélange réactionnel. Cette méthode consiste à sélectionner des états tampons et indicateurs destinés à présenter la même affinité vis-à-vis des protons, de sorte que la quantité relative de tampon protonée est proportionnelle à la quantité d'indicateur protonée, au fur et à mesure que le pH dudit mélange réactionnel évolue. Un mélange réactionnel est ensuite préparé à base d'un tampon, d'un indicateur, d'une hydrolase à tester, et d'un substrat à tester, l'hydrolase étant ensuite amenée à réagir avec ledit substrat. On contrôle ensuite cette réaction en détectant tout changement de la couleur dudit mélange réactionnel, en fonction de l'influence de la réaction sur l'indicateur.
