A method for purifying and recovering biologically active somatotropin monomers from refold solution following the solubilization and naturation of refractile bodies of host cells produced by recombinant DNA methodology. The purification process is based on the discovery that somatotropin monomers and somatotropin oligomers having overlapping isoelectric points may nevertheless be separated by selective precipitation over a narrow pH range. Host cell residues including proteins, pyrogens and other impurities present in the refold solution are effective removed in the process. The purified somatotropin monomers recovered from solution after removing precipitated solids are suitable for parenteral application to target animals without further purification.
Une méthode pour purifier et récupérer des monomères de somatotropine biologiquement active, produite par la technologie de recombinaison de l'ADN, dans une solution de repliement après solubilisation et naturation des corps réfractiles des cellules hôtes. Le procédé de purification est fondé sur la découverte que les monomères de somatotropine et les oligomères de somatotropine ayant des points isoélectriques chevauchant peuvent quand même être séparés par précipitation sélective dans une étendue de pH étroite. Les résidus des cellules hôtes, comprenant les protéines, les pyrogènes et autres impuretés, présents dans la solution de repliement sont éliminés efficacement au cours du procédé. Les monomères de somatotropine purifiés récupérés de la solution après élimination des solides précipités conviennent à une administration par voie parentérale à des animaux cibles sans autre purification.
