Methods of increasing blood flow in a mammalian brain blood vessel characterized by, or otherwise experiencing, decreased blood flow due to an ischemic or other hypoxic event, vasoconstriction or vasospasm following hemorrhagic stroke; due to chronic high blood pressure; and/or due to idiopathic causes are provided. The method for increasing blood flow in such a mammalian brain blood vessel includes administering to a patient in need thereof a therapeutically effective amount of an inhibitor of protein kinase C. In certain embodiments, the inhibitor can be chronically administered without causing desensitization of the patient to the inhibitor. Kits for increasing blood flow in a mammalian brain blood vessel characterized by, or otherwise experiencing, decreased blood flow due to an ischemic or other hypoxic event, vasoconstriction or vasospasm following hemorrhagic stroke; due to chronic high blood pressure; and/or due to idiopathic causes are provided.
La présente invention se rapporte à une méthode permettant d'augmenter le débit sanguin dans un vaisseau sanguin du cerveau d'un mammifère, ledit vaisseau étant caractérisé par un débit sanguin réduit causé par un événement ischémique ou un autre événement hypoxique, une vasoconstriction ou un spasme vasculaire suivant un accident hémorragique, une hypertension chronique, et/ou des facteurs idiopathiques. La méthode permettant d'augmenter le débit sanguin dans un tel vaisseau sanguin du cerveau d'un mammifère selon l'invention consiste à administrer à un patient concerné une dose thérapeutiquement efficace d'un inhibiteur de la protéine kinase C .alpha.. Dans certains modes de réalisation, l'inhibiteur peut être administré de manière chronique sans provoquer de résistance du patient à l'inhibiteur. L'invention concerne aussi des kits permettant d'augmenter le débit sanguin dans un vaisseau sanguin causé par un événement ischémique ou un autre événement hypoxique, une vasoconstriction ou un spasme vasculaire suivant un accident hémorragique, une hypertension chronique, et/ou des facteurs idiopathiques.
