The present invention is directed to stable protein derivatives, e.g., antibodies, antibody fragments or peptides, with at least one free thiol group coupled to N-acetyl-L-cysteine, N-ethyl-maleimide, or cysteine and the methods of making such derivatives. In addition, stable liquid pharmaceutical formulations comprising such proteins or their derivatives and stable lyophilized pharmaceutical formulations comprising such proteins are provided. The present invention is also directed to a method of making a stable Fab' fragment of an antibody and a method of controlling vascularization in injured or cancerous tissue comprising applying to the injured tissue one or more doses of the pharmaceutical formulations.
L'invention concerne des dérivés de protéines stables, par exemple des anticorps, des fragments d'anticorps ou des peptides, avec au moins un groupe thiol libre couplé à N-acétyl-L-cystéine, N-éthyl-maléimide, ou cystéine et des procédés relatifs à l'élaboration de tels dérivés. On décrit aussi des formulations pharmaceutiques liquides stables qui renferment les protéines en question ou leurs dérivés et des formulations pharmaceutiques stables liophylisées renfermant ces protéines. L'invention concerne enfin un procédé d'élaboration de fragment Fab' stable d'anticorps et un procédé de contrôle de vascularisation dans du tissu lésé ou cancéreux, qui consiste à appliquer une ou plusieurs doses de ces formulations au tissu considéré.
