Methods of recombining nucleic acids are provided. In particular, methods for the production of partially double stranded nucleic acids comprising a 3~ overhang from an RNA target and use in methods of recombining polynucleotides is described. These methods do not require thermocycling. The present invention also provides methods of recombining and selection which allow for identification of proteins comprising improved or desired characteristics.

L'invention porte sur des méthodes de recombinaison d'acides nucléiques, et en particulier sur des méthodes d'élaboration d'acides nucléiques partiellement à double brin comprenant un 3' sortant de cible d'ARN, utilisables pour la recombinaison de polynucléotides. Ces méthodes ne nécessitent pas de thermocyclage. L'invention porte également sur des méthodes de recombinaison et de sélection permettant d'identifier des protéines présentant des caractéristiques améliorées ou désirées.
