The invention provides a high throughput device for simultaneously isolating periplasmic fractions from multiple samples of host cells. The device can be formatted to operate in a series. The method includes (a) providing a high throughput device for isolating periplasmic fractions from multiple samples of host cells via attaching a sample chamber to a vacuum chamber and applying a vacuum thereto (b) removing the media with the vacuum and (c) removing the outer membrane of the host cells (d) attaching a collection chamber to the vacuum chamber and applying a vacuum thereto in order to carry out a collection of the periplasmic fractions from the host cells. A workstation is also provided which includes one or more high throughput devices for simultaneously isolating periplasmic space fractions from a plurality of samples of host cells and a device therefore.
L'invention concerne un dispositif à débit élevé permettant d'isoler simultanément des fractions périplasmiques d'échantillons multiples de cellules hôtes. Ce dispositif peut être conçu pour fonctionner en série. Le procédé consiste à utiliser un dispositif à débit élevé destiné à isoler des fractions périplasmiques d'échantillons multiples de cellules hôtes en reliant une chambre d'échantillon à une chambre à vide et à appliquer un vide à ladite chambre, (b) à enlever le milieu avec le vide et (c) à enlever la membrane extérieure des cellules hôtes, (d) à relier une chambre collectrice à la chambre à vide et à appliquer un vide à ladite chambre, de manière à recueillir les fractions périplasmiques des cellules hôtes. L'invention concerne également un poste de travail comprenant un ou plusieurs dispositifs à débit élevé destinés à isoler simultanément des fractions d'espace périplasmiques d'une pluralité d'échantillons de cellules hôtes et elle concerne également ledit dispositif.
