A method for reducing total sample complexity in native or digested biological sample(s), before analysis thereof by mass spectrometry, comprising the following steps: a) selecting a fraction from the entire native or digested biological sample(s) on the basis of pI-value, said fraction comprising native or digested sample representing a subset of or the entire substance population in the sample, said fraction being obtained by e.g. anion exchange chromatography, isoelectric focussing or chromatofocussing; b) separating native or digested sample substances from each other, wherein said separation is by cation exchange chromatography; and c) analysing aid substances by mass spectrometry. The invention also relates to a system for reducing total sample complexity in the above method, comprising a high capacity charge-selective column (anion exchange, isoelectric focussing or chromatofocussing) coupled to a MDLC work flow path comprising a cation exchange column and a RPC column. The system is followed by a MS/MS instrument.
L'invention concerne une méthode pour réduire la complexité totale d'un échantillon dans au moins un échantillon biologique natif ou digéré, avant l'analyse de celui-ci, par une spectrométrie de masse. Cette méthode comprend les étapes consistant à: a) sélectionner une fraction de l'échantillon biologique natif ou digéré, en fonction de la valeur pI, cette fraction comprenant un échantillon natif ou digéré représentant un sous-ensemble de la population de substance de l'échantillon ou représentant la totalité de la population de ces substances, la fraction étant obtenue, par exemple, par une chromatographie par échange anionique, par une focalisation isoélectrique ou par une chromatofocalisation; b) séparer des substances d'échantillon natif ou digéré les unes des autres, cette séparation étant effectuée par une chromatographie par échange cationique; et c) analyse ces substances par une spectrométrie de masse. L'invention concerne un système pour réduire la complexité d'échantillon totale, dans la méthode susmentionnée. Ce système comprend une colonne sélective de charge de capacité élevée (échange anionique, focalisation isoélectrique ou chromatofocalisation) couplée à une trajectoire d'écoulement de travail MDLC comprenant une colonne d'échange cationique et une colonne RPC. Le système de l'invention est associé à un instrument MS/MS.
