The aim of the present invention is to provide a method of acquiring immunological tolerance to a foreign DNA or its expression product whereby the foreign DNA such as a vector carrying a foreign gene incorporated thereinto or its expression product can be recognized not as non-self but as self; a method of sustaining a gene therapeutic effect whereby a rejection to a foreign DNA such as a vector carrying a foreign gene incorporated thereinto or its expression product can be avoided; and a non-human animal which has acquired immunological tolerance to a foreign DNA such as a vector carrying a foreign gene incorporated thereinto or its expression product. A fetal T lymphocytes transferred with a foreign DNA, such as a foreign gene--incorporated viral vector, are introduced into thymus and said foreign DNA is expressed in the thymus organ. The methods of transferring said foreign DNA into a fetal T lymphocyte include, for example, co-cultivating the fetal T lymphocytes with viral vector-infected virus producer cells.
L'invention concerne un procédé d'acquisition de tolérance immunologique à un ADN étranger, tel qu'un vecteur porteur d'un gène étranger intégré dans ce vecteur, ou le produit d'expression de ce vecteur, de façon que l'ADN étranger ou son produit d'expression puisse être reconnu non pas comme "non lui-même" mais comme "lui-même". L'invention concerne également un procédé de prolongation d'un effet thérapeutique génique, permettant d'éviter un rejet d'un ADN étranger, tel qu'un vecteur porteur d'un gène étranger intégré dans le vecteur, ou son produit d'expression, de même qu'un mammifère non humain ayant acquis une tolérance immunologique à un ADN étranger, tel qu'un vecteur porteur d'un gène étranger intégré dans le vecteur, ou son produit d'expression. L'invention concerne encore des lymphocytes T immatures qui possèdent un ADN étranger (par exemple un vecteur de virus porteur d'un gène étranger intégré dans ce vecteur), transféré dans ces lymphocytes, et que l'on introduit dans le thymus, l'ADN étranger ci-dessus s'exprimant alors dans le thymus. On peut transférer l'ADN étranger décrit ci-dessus, dans les lymphocytes T immatures, par exemple, en cocultivant les lymphocytes T immatures avec des cellules productrices de virus et infectées au moyen d'un vecteur de virus.
