The present invention relates to a method for producing a polypeptide comprising using a variant alpha factor signal peptide having a substitution in position 9 resulting in a substitution of an A to either T, S, H, I, F, E, or G. The invention further relate to nucleic acid constructs comprising a first nucleotide sequence encoding the variant signal peptide and a second nucleotide sequence encoding a polypeptide which is foreign to the first nucleotide sequence. Furthermore, it also relates to expression vectors and host cells comprising said nuclei acid construct.
La présente invention concerne un procédé de production d'un polypeptide impliquant d'utiliser un variant de peptide de signalisation de facteur alpha comportant une substitution en position 9, ladite substitution ayant pour effet de remplacer un A par un T, S, H, I, F, E ou G. L'invention concerne également des constructions d'acide nucléique comprenant une première séquence nucléotidique codant pour le variant du peptide de signalisation et une seconde séquence nucléotidique codant pour un polypeptide étranger à ladite première séquence nucléotidique. Enfin, l'invention concerne des vecteurs d'expression et des cellules hôtes comprenant lesdites constructions d'acide nucléique.
