Proteins may be made by genetically engineered microorganisms, the protein being stored in the form of inclusion bodies (IB). The proteins in the inclusion bodies are in an insoluble and inactive form. They may be dissolved using a solubilisation reagent (18), and the resulting solution diluted so that the proteins refold into the active form. This refolding of the protein is enhanced by subjecting a solution or suspension of the protein to low intensity sound waves (25), at a low enough intensity that the protein is not denatured. The intensity may be between 10 and 100 mW/cm2.
On peut fabriquer des protéines par génie génétique sur des micro-organismes, la protéine étant stockée sous la forme de corps d'inclusions (IB). Les protéines dans les corps d'inclusion sont dans un état insoluble et inactif. Elles peuvent être dissoutes au moyen d'un réactif de solubilisation (18), et la solution obtenue diluée de sorte que les protéines se replient en une forme active. Ce repliement de la protéine est amélioré par traitement avec une solution ou une dispersion de la protéine avec des ondes sonores de faible intensité (25), dont l'intensité est suffisamment faible pour que la protéine ne soit pas dénaturée.
